目的观察重组人粒细胞集落刺激因子(rh G-CSF)联合丹参酮ⅡA与丹皮酚配伍对脑缺血大鼠CD34/Brdu(5-溴脱氧尿嘧啶核苷)表达的影响,探讨其治疗脑缺血的机制。方法采用改良线栓法复制脑缺血模型,用免疫组化法检测各组大鼠脑缺血损伤后第14天,缺血侧的皮质和海马区中CD34和Brdu的变化。结果与模型组大鼠比较,假手术组中几乎检测不到CD34和Brdu的表达;rh G-CSF+丹参酮ⅡA+丹皮酚组及rh G-CSF+丹皮酚组的CD34和Brdu阳性细胞计数均有统计学差异。结论 rh G-CSF+丹参酮ⅡA+丹皮酚及rh G-CSF+丹皮酚能促进脑缺血大鼠的血管生成和神经干细胞的增殖。
Objective To observe the effects of rhG-CSF combined with tanshinone ⅡA and paeonol on the expression of CD34 / Brdu (5-bromodeoxyuridine) in rats with cerebral ischemia Ischemia mechanism. Methods The model of cerebral ischemia was established by modified suture method. The changes of CD34 and Brdu in cortex and hippocampus on the 14th day after cerebral ischemia in rats were detected by immunohistochemistry. Results Compared with the model group, the expression of CD34 and Brdu in the sham-operation group was almost undetectable. The counts of CD34 and Brdu positive cells in rhG-CSF + tanshinoneⅡA + paeonol group and rhG-CSF + paeonol group were Statistical differences. Conclusion rhG-CSF + Tanshinone ⅡA + paeonol and rhG-CSF + paeonol can promote angiogenesis and proliferation of neural stem cells in rats with cerebral ischemia.
