目的:探讨亚慢性甲苯染毒对大鼠卵巢的直接毒性作用。方法:将40只雌性大鼠随机分为对照组和甲苯染毒组。对照组给予1 ml不含甲苯的玉米油,染毒组甲苯剂量从低到高分别为35、70、330 mg/kg,连续染毒40~45天,观察甲苯对卵巢脏器系数、组织结构的影响;计数各级卵泡及黄体数目,用TUNEL法检测颗粒细胞凋亡率,并测血清E2、P的浓度。结果:①中、高剂量甲苯组卵巢脏器系数较对照组降低,差异有统计学意义(P<0.05)。②4组之间原始卵泡数目无显著改变,随着染毒剂量的增加,生长卵泡减少,闭锁卵泡增加,黄体数目减少,且中、高剂量组与对照组相比,差异有统计学意义(P<0.05)。③中、高剂量甲苯组颗粒细胞凋亡率(26.6%,35.3%)高于对照组(10.9%),差异有统计学意义(P<0.001)。④中、高剂量甲苯组E2、P较对照组降低,差异有统计学意义(P<0.01)。结论:亚慢性甲苯染毒对大鼠卵巢有毒性作用,且与甲苯剂量呈正相关。
Objective: To investigate the direct toxic effects of sub-chronic toluene exposure on ovary in rats. Methods: Forty female rats were randomly divided into control group and toluene exposure group. The control group was given 1 ml of corn oil without toluene. The dosage of toluene in the exposed group was 35, 70, and 330 mg / kg, respectively, from low to high. The toxicity of toluene to ovary organ coefficient, tissue structure The number of follicles and corpus luteum at each stage was counted. The apoptosis rate of granulosa cells was detected by TUNEL method. The concentrations of E2 and P in serum were also measured. Results: (1) Ovarian organ coefficient of middle and high dose of toluene group was lower than that of control group, the difference was statistically significant (P <0.05). ② There was no significant change in the number of primordial follicles between the four groups. With the increase of exposure dose, the number of growth follicles decreased, the number of atresia follicles increased and the number of corpus luteum decreased. There was significant difference between the medium and high dose groups and the control group (P <0.05). (3) The apoptosis rates of granulosa cells in middle and high dose of toluene group were higher than those in control group (26.6%, 35.3%), the difference was statistically significant (P <0.001). ④ Middle and high dose of toluene group E2, P than the control group decreased, the difference was statistically significant (P <0.01). Conclusion: Subchronic toluene exposure has toxic effects on the ovary of rats and is positively correlated with the dosage of toluene.
