①目的 采用体外实验方法探讨高压氧(HBO)对化疗药物体外诱导急性髓性白血病(AML)细胞Bax和bcl-2基因表达的影响。②方法 高三尖杉酯碱(H)、阿糖胞苷(A)、足叶乙甙(V)3种化疗药物分别作用于AML患者的单个核细胞,一组加HBO,另一组不加HBO,观察各组凋亡细胞、Bax和bcl-2实验前后积分变化、Bax/bcl-2积分比和TUNEL阳性率。同时设无药加HBO组和不加HBO组以排除HBO或其它因素对AMI,细胞的影响。③结果 3种化疗药物加HBO组具有典型凋亡特征的AML细胞明显增多;Bax的表达无明显改变(P>0.05),bcl-2的表达明显减少(P<0.01),Bax/bcl-2积分比明显高于不加HBO组(P<0.05);TUNEL 阳性率明显高于不加HBO组(P<0.01);无药加HBO组与无药不加HBO组实验前后Bax、bcl-2基因的表达无明显改变(P>0.05)。④结论HBO对化疗药物体外促进AML细胞Bax基因的表达无明显影响,但增强了化疗药物对bcl-2基因表达的抑制作用,Bax/bcl-2比值明显增加,因而增强了AML细胞的化疗敏感性,使化疗药物体外诱导AML细胞凋亡增加。单纯HBO对AML细胞Bax和bcl-2基因的表达无明显影响。
Objective To investigate the effect of hyperbaric oxygen (HBO) on Bax and bcl-2 gene expression in acute myeloid leukemia (AML) cells induced by chemotherapeutic drugs in vitro. ② Methods Homoharringtonine (H), cytarabine (A), etoposide (V) three kinds of chemotherapy drugs were treated in AML mononuclear cells, one with HBO, the other without HBO. The apoptotic cells in each group were observed. The integral changes of Bax and bcl-2 before and after the experiment, Bax / bcl-2 integral ratio and TUNEL positive rate were observed. At the same time set no drug plus HBO group and no HBO group to exclude HBO or other factors on AMI, cells. Results The AML cells with typical apoptotic features of three chemotherapeutic drugs plus HBO group were significantly increased, the expression of Bax was not significantly changed (P> 0.05), the expression of bcl-2 was significantly decreased (P <0.01), Bax / bcl-2 (P <0.05). The positive rate of TUNEL was significantly higher than that without HBO (P <0.01). The levels of Bax, bcl-2 before and after treatment with or without HBO No significant changes in gene expression (P> 0.05). Conclusion HBO has no obvious effect on the expression of Bax gene in AML cells induced by chemotherapeutic drugs in vitro, but enhances the inhibitory effect of chemotherapeutic agents on the expression of bcl-2 gene. The ratio of Bax / bcl-2 is significantly increased, which enhances the chemosensitivity of AML cells In addition, chemotherapeutic drugs induce apoptosis of AML cells in vitro. HBO alone had no significant effect on the expression of Bax and bcl-2 genes in AML cells.
