采用后包埋免疫金技术,在透射电镜下识别确定White树脂包埋的结肠上皮细胞和结肠癌细胞内β-葡萄糖醛酸酶超微结构定位,和两者之间比较。结果发现,此酶金标颗粒定位于结肠上皮细胞和结肠癌细胞中的内质网、高尔基复合体和溶酶体中。癌细胞中β-葡萄糖醛酸酶的数量高于正常结肠上皮细胞。提示β-葡萄糖醛酸酶的增多可能与癌细胞旺盛的机能活动和异常的形态结构有关。
Using post-embedding immunogold technology, the ultrastructural localization of β-glucuronidase in colonic epithelial cells embedded in White resin and colon cancer cells was identified by transmission electron microscopy and compared. As a result, it was found that the gold particles of this enzyme were localized in the endoplasmic reticulum, Golgi complex and lysosome in colonic epithelial cells and colon cancer cells. The number of β-glucuronidase in cancer cells is higher than that in normal colon epithelial cells. It suggests that the increase of β-glucuronidase may be related to the exuberant function of cancer cells and abnormal morphological structure.
