背景:人类对脑出血的研究常采用大鼠的脑出血模型,但脑出血模型中缺乏一些重要指标描述。目的:为脑出血的发病机制、治疗等研究提供有效可靠的疾病动物模型及相关指标。设计:随机对照实验研究。地点和对象:成年SD大鼠50只,均为雄性,实验在四川大学基础与法医学院人体解剖学教研室完成。干预:SD大鼠50只随机分为自体血注入组(20只)和胶原酶注入组(20只),在大鼠尾状核分别注入自体血50μL和胶原酶5.001nkat,建立两种不同的脑出血模型,术后3d自体血注入组大鼠死亡5只,胶原酶注入组死亡共6只。主要观察指标:两组大鼠神经功能学评分,病理变化,血肿大小,免疫组织化学染色结果。结果:血肿大小犤自体血注入组为(10.65±3.12)μL、胶原酶注入组为(40.26±1.45)μL犦,两者比较差异有统计学意义(t=33.102,P<0.001)、阳性小胶质犤自体血注入组为(32.6±3.6)个、胶原酶注入组为(8.8±2.4)个犦,两者比较差异有统计学意义(t=20.885,P<0.001)。结论:由于两种方法建立的脑出血模型各种指标不同,因此可以根据不同的研究目的采用不同的动物模型。
BACKGROUND: Human studies on intracerebral hemorrhage often use the rat model of intracerebral hemorrhage, but some important indications are lacking in the cerebral hemorrhage model. Objective: To provide an effective and reliable disease animal model and related indicators for the study of the pathogenesis and treatment of cerebral hemorrhage. Design: Randomized controlled trial. Place and object: 50 adult male Sprague-Dawley rats, all male, were completed in the Department of Human Anatomy of the Department of Basic and Forensic Medicine, Sichuan University. Intervention: 50 SD rats were randomly divided into autologous blood injection group (20) and collagenase injection group (20). In the caudate nucleus, 50 μL of autologous blood and 5.001nkat of collagenase were respectively injected to establish two different After intracerebral hemorrhage, there were 5 dead rats injected with autologous blood and 6 dead rats injected with collagenase. MAIN OUTCOME MEASURES: Neurological scores, pathological changes, hematoma size and immunohistochemistry results of two groups of rats. Results: The hematoma size was significantly higher in the autoimmune blood injection group than that in the autoimmune group (40.66 ± 1.45) μL (10.65 ± 3.12) μL, the difference was statistically significant (t = 33.102, P <0.001) There were significant differences between the two groups (t = 20.885, P <0.001) in the group of glial injection of autologous blood (32.6 ± 3.6) and the group of collagenase injection (8.8 ± 2.4). CONCLUSIONS: Since the various models of cerebral hemorrhage established by the two methods are different, different animal models may be used for different research purposes.
