目的探讨吸烟对大鼠睾丸结构及睾丸波形蛋白表达、血浆中抑制素B含量的影响。方法将清洁级成年雄性SD大鼠160只,随机分为对照组和低(10只香烟)、中(20支香香烟)和高(30只香烟)实验组,每组10只。实验组大鼠置于自制染毒箱中,采用静式染毒方法,分别染毒2、4、6、8、12周,每日1次,连续染毒。观察大鼠睾丸组织形态结构,波形蛋白表达,测定血浆中抑制素B的含量。结果实验组低、中、高剂量组脏器系数、血浆中抑制素B含量低于空白对照组(P<0.05),且随着染毒时间的延长,呈降低趋势;免疫组化结果显示,染毒组波形蛋白阳性表达减少,其表达量较空白组降低(P<0.05);光学显微镜观察提示实验组睾丸曲细精管及生精细胞受到不同程度的损伤。结论香烟烟雾中的有害成分影响睾丸组织支持细胞的结构和功能损害。
Objective To investigate the effects of smoking on testicular structure, testis vimentin and plasma inhibin B levels in rats. Methods 160 adult male SD rats of clean grade were randomly divided into control group and low (10 cigarettes), medium (20 cigarettes) and high (30 cigarettes) experimental groups, 10 in each group. The rats in experiment group were placed in self-made poisoning cases and were exposed to 2,4,6,8,12 weeks, once a day for continuous exposure. The morphology and structure of testis and the expression of vimentin were observed. The content of inhibin B in plasma was measured. Results In the low, medium and high dose groups, the organ coefficient and the content of inhibin B in plasma were lower than those in the blank control group (P <0.05), and decreased with the prolongation of exposure time. The results of immunohistochemistry showed that, Compared with the blank group, the expression of vimentin in the exposure group decreased significantly (P <0.05). The optical microscope observation indicated that the testicular seminiferous tubules and spermatogenic cells in the experimental group were damaged to some extent. Conclusion The harmful components of cigarette smoke affect the structure and function damage of testicular tissue supporting cells.
