目的:探讨分泌型磷脂酶A2(sPLA2)和环氧合酶-2(COX-2)在早产产妇胎膜中的表达及其与早产发生的关系。方法:采用实时荧光定量PCR技术(RT-PCR)检测8例早产产妇(早产临产组)、12例足月临产产妇(足月临产组)和12例足月未临产产妇(对照组)胎膜中sPLA2和COX-2 mRNA的表达水平(以目的基因/内参基因×100表示)。结果:sPLA2和COX-2 mRNA在早产临产组胎膜中的表达水平分别为(159.51±38.97)和(144.17±57.31),在足月临产组胎膜中的表达水平分别为(135.07±25.94)和(172.12±66.29),在对照组胎膜中的表达水平分别为(91.03±33.68)和(84.25±42.78)。早产临产组及足月临产组明显高于对照组,差异有统计学意义(P<0.05);早产临产组与足月临产组比较,差异无统计学意义(P>0.05)。3组胎膜中sPLA2和COX-2的表达水平呈正相关,相关系数γ=0.413。结论:sPLA2和COX-2可能参与早产分娩的发动,在早产的发生中起一定作用。
Objective: To investigate the expression of secreted phospholipase A2 (sPLA2) and cyclooxygenase-2 (COX-2) in the fetal membranes of prematurity women and its relationship with preterm birth. Methods: The fetal membranes of 8 preterm labor (premature labor), 12 full term labor (full term labor) and 12 full term nonpregnant mothers (control) were detected by real-time fluorescence quantitative PCR (RT- The expression levels of sPLA2 and COX-2 mRNA (indicated by the gene of interest / reference gene × 100). Results: The expression levels of sPLA2 and COX-2 mRNA in preterm delivery group were (159.51 ± 38.97) and (144.17 ± 57.31), respectively. The expression levels of sPLA2 and COX-2 mRNA in preterm labor group were (135.07 ± 25.94) And (172.12 ± 66.29), respectively. The expression levels in the control group were (91.03 ± 33.68) and (84.25 ± 42.78), respectively. The preterm delivery group and term delivery group were significantly higher than the control group, the difference was statistically significant (P <0.05); preterm delivery group and full-term labor group, the difference was not statistically significant (P> 0.05). The expression levels of sPLA2 and COX-2 in the three groups of membranes were positively correlated, the correlation coefficient γ = 0.413. Conclusion: sPLA2 and COX-2 may participate in the onset of premature labor and play a role in the occurrence of premature labor.
