目的:建立葫芦素混合胶束中药物含量及包封率测定的反相高效液相(RP-HPLC)法。方法:采用DiamonsilTMC18色谱柱(200 mm×4.6 mm,5μm);流动相为乙腈-水-磷酸(45∶55∶0.1);柱温:室温;流速:1.0 mL.min-1;检测波长:228 nm。结果:葫芦素B与辅料及溶剂的色谱峰分离良好,葫芦素B在0.25~20μg.mL-1的浓度范围内,峰面积与浓度的线性关系良好(r=0.999 9,n=7),回收率在98.8%~100.9%之间,方法的精密度良好。结论:该方法准确可靠,简单快速,可用于葫芦素混合胶束含量及包封率的测定。
Objective: To establish a reversed-phase high performance liquid chromatography (RP-HPLC) method for the determination of the drug content and entrapment efficiency of cucurbitacin mixed micelles. METHODS: Diamonsil TMMC18 column (200 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile-water-phosphoric acid (45:55:0.1). The column temperature was at room temperature and the flow rate was 1.0 mL.min-1. nm. Results: The calibration curve of cucurbitacin B with its excipients and solvents was well separated. The linear relationship between peak area and concentration of cucurbitacin B was good (r = 0.999 9, n = 7) in the range of 0.25 ~ 20μg.mL-1 The recovery rate is between 98.8% and 100.9%, and the precision of the method is good. Conclusion: The method is accurate, reliable, simple and rapid and can be used for the determination of cucurbitacin mixed micelle content and entrapment efficiency.
