用染料探针对蛋白质的定量已应用于临床医学及生命科学的研究中,但染料以金属配合物的形式对蛋白质的定量及其机理研究较少。本文首次报道,在pH25的ClarkLubs缓冲介质中,用氯磺酚SCu(Ⅱ)配合物与BSA作用,生成深蓝色的三元复合物,该复合物的λmax为724nm,比其金属配合物的λmax红移53nm,比试剂本身红移160nm,比氯磺酚SBSA的二元复合物红移93nm。研究了最佳反应条件,制作了几种蛋白质的工作曲线。测定的线性范围宽,灵敏度高。用于实际人血清样品的测定干扰少,准确度高。
Quantitative protein quantification with dye probes has been used in clinical and life science research, but dyes have little research on the quantification of proteins and their mechanisms in the form of metal complexes. It was first reported in this paper that a dark blue ternary complex was formed using the SCu (Ⅱ) complex with chlorosulfonated SCu (Ⅱ) complex in Clark Lubs buffer medium at pH25. The λmax of this complex was 724 nm, which was higher than that of its metal complex Redshift 53 nm, red-shifted 160 nm from the reagent itself, red-shifted 93 nm from the binary complex of sulfosalicylic SBSA. The optimum reaction conditions were studied and the working curves of several proteins were prepared. Determination of the linear range, high sensitivity.
