目的研究深圳福田地区不同来源的副溶血性弧菌的病原学特征、毒力基因、血清型及分子分型特征情况。方法对本地区2007年-2014年107株不同来源的副溶血性弧菌进行血清型分型;采用荧光PCR方法对全部副溶血性弧菌进行tdh、trh毒力基因检测;采用脉冲场凝胶电泳技术对其进行PFGE分型,并用Bio Numerics软件进行聚类分析。结果 107株菌株共得到22种不同血清型,主要血清型为O3∶K6、O4∶K8、O2∶K28、O4∶K13、O1∶K25、O4∶K34和O5∶K68等。主要毒力基因为tdh+trh-,其次为tdh-trh-。采用PFGE分型共得到58种不同电泳图谱,主要的图谱类型为K16O3K6N11.FT0001、K16O3K6N11.FT0009、K16O3K6N11.FT0008、K16O3K6N11.FT0002和K16O4K8N11.FT00-03。结论主要血清型菌株所携带的毒力基因一致,主要为tdh+trh-,其次为tdh-trh-;血清型O3∶K6和O4∶K8菌株被PFGE聚类分型的效力最高,分别分出19和11种亚型条带;血清型O1∶K25、O5∶K68和O11∶K36PFGE条带图谱与O3∶K6图谱相似,均携带tdh+trh-毒力基因,遗传关系接近,可能为O3∶K6的血清变异型。
Objective To study the etiological characteristics, virulence genes, serotypes and molecular typing of Vibrio parahaemolyticus from different origins in Futian, Shenzhen. Methods 107 isolates of Vibrio parahaemolyticus from 2007 to 2014 were genotyped in this region. The virulence genes tdh and trh of all Vibrio parahaemolyticus were detected by fluorescence PCR. The pulsed-field gel electrophoresis PFGE genotyping and Bio Numerics software clustering analysis. Results A total of 22 different serotypes were obtained from 107 isolates. The major serotypes were O3: K6, O4: K8, O2: K28, O4: K13, O1: K25, O4: K34 and O5: K68. The major virulence genes are tdh + trh-, followed by tdh-trh-. A total of 58 different electropherograms were obtained by PFGE typing. The main patterns were K16O3K6N11.FT0001, K16O3K6N11.FT0009, K16O3K6N11.FT0008, K16O3K6N11.FT0002 and K16O4K8N11.FT00-03. Conclusions The major serotype strains carry the same virulence genes, mainly tdh + trh-, followed by tdh-trh-; serotypes O3: K6 and O4: K8 strains are the most potent by PFGE clustering, 19 and 11 subtype bands. The serotypes O1: K25, O5: K68 and O11: K36PFGE bands were similar to those of O3: K6, all carrying the tdh + trh- virulence genes. K6 serum variant.
