目的:建立一种快速测定盾叶薯蓣中伪原薯蓣皂苷含量的方法。方法:首先扫描76份盾叶薯蓣样本的近红外光谱(NIRS)图,通过对光谱预处理方法及建模波段的考察,选择标准归一化+一阶导数作为最优光谱预处理方法,10118.36~4007.37cm-1为建模最佳波段。在此基础上,以高效液相色谱(HPLC)法测定的伪原薯蓣皂苷的含量为参考值,结合偏最小二乘法(PLS)建立NIRS与HPLC分析值之间的定量分析模型。结果:所建模型具有良好的预测能力(r=0.97596),内部交叉验证均方差为0.16975,预测均方差为0.0896。结论:利用NIRS技术可以快速、准确地测定盾叶薯蓣中伪原薯蓣皂苷的含量。
Objective: To establish a rapid determination of Dioscorea zingiberensis pseudo-original method of diosgenin content. Methods: Near infrared spectroscopy (NIRS) images of 76 samples of Dioscorea zingiberensis were screened. Based on the methods of spectral pretreatment and modeling, the standard normalization + first order derivative was selected as the optimal pretreatment method. 10,118.36 ~ 4007.37cm-1 for modeling the best band. On this basis, the quantitative analysis model between NIRS and HPLC analysis was established by PLS method with reference to the content of pseudo-original diosgenin determined by high performance liquid chromatography (HPLC). Results: The model has good predictive ability (r = 0.97596), internal cross-validation mean square deviation 0.16975, and prediction mean square deviation 0.0896. Conclusion: The content of pseudo-yam saponins in Dioscorea zingiberensis CHWright can be determined quickly and accurately by using NIRS technique.
