目的:蛇六谷乙酸乙酯萃取物采用硅胶柱层析后的所得组分进行体外抗肿瘤活性筛选。方法:使用硅胶吸附层析分离技术,对蛇六谷乙酸乙酯萃取物进行化学成分分离;应用MTT法检测各组分对SGC-7901、HepG-2及C6细胞增殖的影响。结果:蛇六谷乙酸乙酯萃取物经硅胶柱层析分离,得到A1~A6,B1~B4,C,D等12个组分;MTT法实验显示A1、A2组分对SGC-7901、HepG-2及C6细胞的增殖均具有较强的抑制作用,并呈一定的量效关系。其中A1和A2在20μg浓度时,对HepG-2、SGC-7901、C6细胞增殖的抑制率分别达66.8%和68.4%;67.5%和77.8%;73.3%和70.6%。结论:蛇六谷乙酸乙酯萃取物经硅胶柱层析分离得12个组分中以A1、A2组分的抑瘤作用较强,为蛇六谷最佳有效部位。
OBJECTIVE: To screen the anti-tumor activity of the extract from ethylhexyl glutamate in ethyl acetate extract by silica gel column chromatography. Methods: Silica gel adsorption chromatography was used to separate the ethyl acetate extracts from snake six leaves. The effects of different components on the proliferation of SGC-7901, HepG-2 and C6 cells were detected by MTT assay. Results: Twelve components (A1 ~ A6, B1 ~ B4, C, D, etc.) were obtained by chromatography on silica gel column. The results of MTT assay showed that the activities of A1 and A2 on SGC-7901, HepG -2 and C6 cells proliferation have a strong inhibitory effect, and showed a certain dose-effect relationship. Among them, A1 and A2 inhibited the proliferation of HepG-2, SGC-7901 and C6 cells by 66.8% and 68.4%, 67.5% and 77.8%, 73.3% and 70.6%, respectively. CONCLUSION: The components of A1 and A2 of 12 components isolated from the extract of ethylhexyl glutamate in ethyl acetate extract by silica gel column are more effective in inhibiting tumor, which is the best effective site of snake Liu Valley.
