目的探讨哮喘豚鼠外周血淋巴细胞凋亡及白介素- 5(IL- 5)、粒细胞 /巨噬细胞集落刺激因子 (GM- CSF)mRNA的表达。方法实验分为哮喘组和对照组,各 15只豚鼠。采用原位末端标记 (TUNEL)法检测豚鼠外周血淋巴细胞 IL- 5、 GM- CSFmRNA的表达。结果哮喘组外周血淋巴细胞凋亡率为 (3.50± 1.00)%,对照组为 (6.80± 1.50)%,哮喘组显著低于对照组 (P< 0.01)。哮喘组 IL- 5、 CM- CSFmRNA表达阳性率分别为 (58.00± 6.00)%和 (49.00± 4.00)%,对照组分别为 (30.00± 2.00)%和 (24.00± 2.00)%,哮喘组显著高于对照组。结论哮喘豚鼠外周血淋巴细胞 IL- 5、 GM- CSFmRNA的高表达,可能与淋巴细胞被激活、凋亡受抑制,从而分泌细胞因子增加有关。
Objective To investigate the apoptosis of peripheral blood lymphocytes and the expression of interleukin - 5 (IL - 5) and granulocyte / macrophage colony stimulating factor (GM - CSF) mRNA in asthmatic guinea pigs. Methods The experiment was divided into asthma group and control group, with 15 guinea pigs each. TUNEL method was used to detect the expression of IL-5 and GM-CSF mRNA in peripheral blood of guinea pigs. Results The apoptotic rates of peripheral blood lymphocytes in asthma group were (3.50 ± 1.00)% and (6.80 ± 1.50)% respectively in asthma group and significantly lower than those in control group (P <0.01). The positive rates of IL-5 and CM-CSF mRNA expression in asthma group were (58.00 ± 6.00)% and (49.00 ± 4.00)% respectively, while those in control group were (30.00 ± 2.00)% and In the control group. Conclusion The overexpression of IL-5 and GM-CSF mRNA in peripheral blood lymphocytes of asthmatic guinea pigs may be related to the activation of lymphocytes, the inhibition of apoptosis, and the increased secretion of cytokines.
