本文报道用HPLC法测定血浆或制剂中的氟尿嘧啶(5-FU),以胞啶为内标,该方法快速、精密.5-FU和内标的理论塔板数分别为242和208,其分离度为4.65,测定与5-FU联合化疗的其他抗癌药物均不干扰,用于制剂分析相对标准偏差<3.5%.方法:HPLC法,流动相为0.025mol/L正己烷磺酸水溶液,流速4ml/min,色谱柱内装10μm8mm×10cm MB~(TM) C_(??),检测波长为270nm.血浆中5-FU标准曲线:由于5-FU血浆中存在浓度范围宽,所以其标准曲线0.025~1.0μg/ml用于低浓度范围,1~25μg/ml用于高浓度范围.取空白血浆1ml,加适量0.1μg/ml或0.02μg/ml的5-FU水溶液,在上述浓度范围内使呈不同浓度.加色谱级纯
