[目的]探讨Nucleostemin(NS)基因特异性RNA干扰对食管癌Eca-109细胞株增殖和凋亡情况的影响。[方法]用NS特异性siRNA表达载体转染食管癌Eca-109细胞株,利用MTT法测试细胞增殖抑制率;RT-PCR法检测NS基因表达量的变化,流式细胞仪法检测细胞周期分布和细胞凋亡情况。[结果]与阴性对照组相比,PCDNA4/C-NS-silencer组细胞的增殖速率降低,24h、48h、72h细胞增殖抑制率分别为57.8%、74.9%、87.3%;NS基因表达量下降,G0/G1期细胞百分率显著升高,S期细胞减少,细胞凋亡率增加。[结论]RNAi技术可显著抑制食管癌Eca-109细胞株NS基因表达,导致细胞增殖缓慢,细胞周期阻滞,凋亡增加。
[Objective] To investigate the effect of Nucleostemin (NS) gene-specific RNA interference on the proliferation and apoptosis of esophageal cancer Eca-109 cell line. [Methods] The esophageal cancer cell line Eca-109 was transfected with NS-specific siRNA expression vector and the cell proliferation inhibition rate was measured by MTT assay. The expression of NS gene was detected by RT-PCR and the cell cycle distribution was detected by flow cytometry And apoptosis. [Results] Compared with the negative control group, the proliferation rate of PCDNA4 / C-NS-silencer group was decreased, and the inhibition rates of proliferation were 57.8%, 74.9% and 87.3% at 24h, 48h and 72h respectively. The NS gene expression decreased, G0 / G1 phase cells was significantly increased, S phase cells decreased, the rate of apoptosis increased. [Conclusion] RNAi can significantly inhibit NS gene expression in esophageal cancer Eca-109 cell line, resulting in slow cell proliferation, cell cycle arrest and apoptosis.
