目的探讨外周血干扰素调节因子4(IRF4)、辅助性T细胞17(Th17)细胞及其相关细胞因子与梅毒血清固定形成的关联性。方法选取本院2014年4月至2016年7月收集的梅毒血清固定患者血清标本69例作为研究组,另选同期健康体检者血清标本67例作为对照组。各组标本均予以分离外周血单个核细胞处理,同时实施外周血单个核细胞IRF4、Th17、CD4细胞检测,应用流式细胞术检测Th17及IRF4细胞表达情况。分析比较2组外周血单个核细胞IRF4、Th17、CD4细胞的表达情况及Th17、IRF4细胞的表达水平。结果研究组外周血单个核细胞IRF4细胞表达水平与对照组比较差异无统计学意义(P>0.05);研究组外周血单个核细胞Th17、CD4细胞表达水平低于对照组,差异有统计学意义(P<0.05);研究组Th17占CD4细胞比例、CD4+IRF4+T细胞占CD4细胞比例、CD4+IL-17+IRF4+T细胞占CD4+IL-17+T细胞比例均低于对照组,差异有统计学意义(P<0.05)。结论梅毒血清固定患者外周血中Th17、CD4细胞表达显著低于正常水平,Th17细胞及其相关细胞因子直接参与梅毒血清固定的形成;梅毒血清固定患者外周血中IRF4细胞表达水平与正常人群无明显差异,IRF4通过对Th17细胞产生作用而间接参与梅毒血清固定。
Objective To investigate the relationship between IRF4, Th17 cells and related cytokines in the formation of syphilis serum. Methods Sixty-nine cases of sera from patients with syphilis sera collected from April 2014 to July 2016 in our hospital were selected as the study group, and 67 cases of sera from the same period were selected as the control group. Peripheral blood mononuclear cells (PBMCs) were isolated from all the groups. At the same time, IRF4, Th17 and CD4 were detected in peripheral blood mononuclear cells. The expression of Th17 and IRF4 cells was detected by flow cytometry. The expression of IRF4, Th17 and CD4 cells in peripheral blood mononuclear cells and the expression of Th17 and IRF4 cells in the two groups were analyzed. Results The expression level of IRF4 in peripheral blood mononuclear cells in the study group was not significantly different from that in the control group (P> 0.05). The levels of Th17 and CD4 in peripheral blood mononuclear cells in the study group were lower than those in the control group (P <0.05). The proportion of Th17 cells to CD4 cells, CD4 + IRF4 + T cells and CD4 + IL-17 + IRF4 + T cells in the study group were lower than those in the control group , The difference was statistically significant (P <0.05). Conclusion The expression of Th17 and CD4 in peripheral blood of patients with syphilis serum is significantly lower than normal, and Th17 cells and related cytokines are directly involved in the formation of syphilis serum. The expression level of IRF4 in peripheral blood of patients with syphilis serum is not significantly higher than that in normal subjects Differentially, IRF4 indirectly participates in syphilis sera fixation by acting on Th17 cells.
