目的检测长链非编码RNA UCA1在肾癌组织、肾癌细胞系中的表达水平,比较配对组织间、细胞系之间的表达差异,分析UCA1表达水平与患者临床特征之间的关系,探究其在肾癌中的临床意义。方法提取肾癌细胞系及46对配对组织中的总RNA,反转录获得c DNA后通过实时荧光定量PCR方法检测UCA1的表达量,使用统计学软件分析UCA1表达的差异性及其表达水平与患者临床特征之间的相关性。结果 UCA1在肾癌组织中表达水平明显高于配对癌旁正常组织(P<0.001),在肾癌细胞(786-O、ACHN、769P、Caki-2)中表达水平高于人胚肾细胞(293T)(P<0.001)。标本中共有38例(82.609%)肾癌标本UCA1表达上调,癌组织中UCA1表达量与患者临床特征无明显相关性(P>0.05)。结论 UCA1在肾癌组织及细胞系中表达明显上调,提示其和肾癌的发生发展有一定关系。
Objective To detect the expression of UCA1, a long-chain non-coding RNA, in renal cell carcinoma and renal cell carcinoma cell lines, compare the expression differences between paired tissues and cell lines, analyze the relationship between UCA1 expression level and clinical features of patients, In renal cell carcinoma clinical significance. Methods The total RNA was extracted from renal cell carcinoma cell lines and 46 pairs of matched tissues. Reverse transcription of c DNA was performed to detect the expression of UCA1 by real-time fluorescence quantitative PCR. Statistical analysis was used to analyze the difference of UCA1 expression and its expression level with Correlation between patient clinical characteristics. Results The expression level of UCA1 in renal cell carcinoma was significantly higher than that in matched normal tissues (P <0.001), and higher in renal carcinoma cells (786-O, ACHN, 769P and Caki-2) than in human embryonic kidney cells 293T) (P <0.001). There were 38 cases (82.609%) of renal cancer specimens UCA1 expression was up-regulated, the expression of UCA1 in cancerous tissues had no significant correlation with the clinical features (P> 0.05). Conclusion UCA1 expression in renal cell carcinoma and cell lines was significantly increased, suggesting that it has a certain relationship with the occurrence and development of renal cell carcinoma.
