目的:观察肺岩宁方对Lewis肺癌小鼠移植瘤VEGF及其受体Flt-1、KDR蛋白表达的影响。方法:采用C57BL/6小鼠,接种Lewis肺癌细胞株,随机分为对照组、化疗组、肺岩宁组和综合组,在接种后5天肿瘤直径约为5 mm时,开始给予0.9%NaCl溶液、DDP、肺岩宁、DDP+肺岩宁治疗。用药21 d,第22天处死小鼠。观察各组肺转移灶抑制率,免疫组化测定肿瘤VEGF及其受体Flt-1、KDR蛋白表达。结果:与对照组相比,肺岩宁组及综合组能显著降低肺转移灶发生率(P<0.05)。免疫组化结果显示:肺岩宁方能够显著降低肿瘤VEGF及其受体KDR水平。肺岩宁方能够减少肿瘤Flt-1表达,但无统计学意义。结论:益气养精、解毒散结的肺岩宁方具有明显的抗肺癌侵袭转移作用。肺岩宁方可以干扰VEGF与其受体(主要是KDR)的相互作用,从而抑制VEGF的生物活性,这可能是肺岩宁方抗肺癌侵袭转移的机制之一。
Objective: To observe the effect of Feiyanning Decoction on the expression of VEGF and its receptor Flt-1, KDR protein in transplanted Lewis lung cancer in mice. Methods: Lewis lung carcinoma cell lines were inoculated with C57BL / 6 mice and were randomly divided into control group, chemotherapy group, Feiyanning group and combination group. At 5 days after inoculation, tumor diameter was about 5 mm, 0.9% NaCl Solution, DDP, lung Yan Ning, DDP + Feiyanning treatment. On the 21st day, the mice were sacrificed on the 22nd day. The inhibitory rate of lung metastasis in each group was observed, and the expression of VEGF and its receptor Flt-1 and KDR protein were detected by immunohistochemistry. Results: Compared with the control group, the Feiyanning group and the combination group could significantly reduce the incidence of lung metastases (P <0.05). The results of immunohistochemistry showed that “Feiyanning Decoction” can significantly reduce the level of VEGF and its receptor KDR. Feiyanningfang can reduce tumor Flt-1 expression, but not statistically significant. Conclusion: Feiyan nourishing, detoxification Sanjie lung Yanning Fang has obvious anti-invasion and metastasis of lung cancer. Feiyanning can interfere with the interaction between VEGF and its receptor (mainly KDR) and thus inhibit the biological activity of VEGF, which may be one of the mechanisms of Feiyanning on lung cancer invasion and metastasis.
