烟草花叶病毒(TMV)的弱毒株SA-10是用变温处理心叶烟(Nicotiana glutihosa)病株而得到的。其诱变程序:常温(25℃)和高温(30—35℃)交替处理病株返复十次。以“三生”烟(N.tabacum CV.Samsun)上的症状轻重来鉴别致弱程度,以心叶烟叶碟上的枯斑数测定病毒的浓度。SA-10弱毒株在“三生”烟上无症状,增殖浓度低于烟草花叶病毒普通株(TMV-O),失毒温度90℃,稀释限点10~(-7)—10~(-8),血清反应与TMV-O相同。可干扰TMV-O的增殖。
The attenuated strain SA-10 of tobacco mosaic virus (TMV) was obtained by variable temperature treatment of the Nicotiana glutiocosa strain. The mutagenesis program: normal temperature (25 ℃) and high temperature (30-35 ℃) alternate treatment of diseased plants ten times. The severity of symptoms was identified by the severity of symptoms on N. tabacum CV.Samsun, and the virus concentration was determined by the number of dead spots on the leaf discs of the heart-leaf tobacco. The attenuated strain of SA-10 was asymptomatic in “SanSheng” tobacco and the proliferation concentration was lower than that of TMV-O. The detoxification temperature was 90 ℃ and the dilution limit was 10 ~ (-7) -10 ~ ( -8), the same as seroconversion with TMV-O. Can interfere with the proliferation of TMV-O.
