目的 研究急性肺损伤(ALI)对肺组织β-防御素-2基因表达的影响。方法 将60只雄性健康SD大鼠随机分成两组:对照组和急性肺损伤组,每组30只。5%苯巴比妥钠 120mg/kg腹腔注射麻醉,以LPS 200 μg/kg尾静脉注射复制急性肺损伤模型;对照组用等量生理盐水(NS)尾静脉注射。于尾静脉注射LPS或NS后0h、6h、12h、1d、3d、5d时杀死大鼠,剖胸取出肺组织。采用Trizol一步法提取肺组织总RNA,以GAPDH为内参,半定量RT-PCR检测肺组织β-防御素-2基因的表达水平,双脱氧末端终止法测序鉴定PCR产物的特异性。结果 对照组0h时肺组织中β-防御素-2基因表达水平为 0.77± 0.14,各时间点肺组织中β-防御素-2基因表达水平无显著性变化(P>0.05)。ALI组6h时肺组织β-防御素-2基因的表达水平与对照组相比无显著性差异(P>0.05),而12h、1d、3d、5d时显著增高(P<0.05)。结论 β-防御素-2基因在大鼠肺组织中有组成型表达,ALI上调肺组织中β防御素-2基因的表达。β-防御素-2基因的诱导性表达可能与ALI的发生发展有关。
Objective To study the effect of acute lung injury (ALI) on the expression of β-defensin-2 gene in lung tissue. Methods Sixty male SD rats were randomly divided into two groups: control group and acute lung injury group, 30 rats in each group. 5% phenobarbital sodium 120mg / kg by intraperitoneal injection of anesthesia, LPS 200μg / kg tail vein injection of acute lung injury model; control group with the same amount of saline (NS) tail vein injection. Rats were sacrificed at 0h, 6h, 12h, 1d, 3d, 5d after tail vein injection of LPS or NS, and lung tissue was removed by thoracotomy. Total RNA was extracted from the lung tissue by one-step Trizol method. The expression of β-defensin-2 gene in lung tissue was detected by semi-quantitative RT-PCR with GAPDH as an internal control. The specificity of the PCR product was identified by dideoxy terminator sequencing. Results The expression of β-defensin-2 gene in lung tissue at 0h was 0.77 ± 0.14 in the control group, and there was no significant change in the expression of β-defensin-2 gene at each time point (P> 0.05). The level of β-defensin-2 expression in lung tissue of ALI group at 6h was not significantly different from that in control group (P> 0.05), but significantly increased at 12h, 1d, 3d and 5d (P <0.05). Conclusion The β-defensin-2 gene is constitutively expressed in rat lung tissue. ALI up-regulated the expression of β-defensin-2 gene in lung tissue. Inducible expression of β-defensin-2 gene may be related to the occurrence and development of ALI.
