[目的]研究蛋白激酶C(PKC)信号途径在苯并(a)芘 (BaP)影响热休克蛋白70(HSP70)基因表达中的作用。[方法]离体培养猪主动脉内皮细胞 ,不同浓度BaP(0、0.1、0.5、1、5、10μmol/L)染毒24h,并以12_十四酸佛波酯_13_乙酸盐 (PMA)和抑制剂Staurosporine于不同时间点进行干预 ,以Western__blot法检测各组细胞HSP70表达的变化。 [结果]BaP抑制HSP70表达 ;PKC的激动剂 (PMA)可完全或部分消除BaP对HSP70的抑制作用 ,而这种作用可迅速被PKC的抑制剂(Staurosporine ,STP)所阻断。[结论]BaP可能通过抑制内皮细胞PKC活性而抑制HSP70的表达
[Objective] To investigate the role of protein kinase C (PKC) signal pathway in the gene expression of heat shock protein 70 (HSP70) induced by benzo (a) pyrene (BaP). [Method] The porcine aortic endothelial cells were cultured in vitro and incubated with different concentrations of BaP (0, 0.1, 0.5, 1, 5 and 10 μmol / L) for 24 h. Phosphatidyl_13_ acetate (PMA) and inhibitor Staurosporine at different time points. Western blotting was used to detect the expression of HSP70 in each group. [Results] BaP inhibited the expression of HSP70; PKC agonist (PMA) completely or partially abolished the inhibitory effect of BaP on HSP70, which was rapidly blocked by PKC inhibitor (Staurosporine, STP). [Conclusion] BaP may inhibit the expression of HSP70 by inhibiting the PKC activity of endothelial cells
