目的在倒置显微镜下观察转导及未转导TNF-α基因DNL抗人舌鳞癌Tca8113细胞的过程。方法在24孔板内Tca8113细胞贴壁后加入DNL,定期观察。结果12h转导及未转导基因组相同,肿瘤细胞被大量DNL包围;48时转导基因组已几乎见不到肿瘤细胞,未转导基因组肿瘤细胞虽减少,但仍可见很多完整的肿瘤细胞。结论转导TNF-α基因能够提高DNL的抗肿瘤活性,提示转导基因DNL抗肿瘤的机制可能是接触杀伤。
Objective To observe the process of transduction and non-transduction of DNL anti-human tongue squamous cell carcinoma Tca8113 cells under inverted microscope. Methods DNL was added into Tca8113 cells in 24-well plates and observed regularly. Results The transduced and non-transduced genomes were the same at 12h. The tumor cells were surrounded by a large number of DNLs. At 48h, almost no tumor cells were seen in the transduced genomes, but fewer intact tumor cells were seen in the untransformed genomes. Conclusion Transduction of TNF-α gene can increase the antitumor activity of DNL, suggesting that the mechanism of DNL antitumor may be contact killing.
