目的观察血管内皮抑素(Endostatin)对小鼠体内Lewis肺癌生长、血管生成及转移的影响。方法将荷Lew is肺癌的C57鼠进行不同剂量组的内皮抑素血管内皮抑素和顺铂干预,观察肿瘤生长、移植瘤及转移瘤体内的微血管密度(MVD)的表达及转移发生率,采用SPSS19.0进行统计学分析。结果血管内皮抑素对鼠Lewis肺癌的生长有明显的抑制作用,并能显著下调移植瘤及转移瘤内微血管密度,减少肿瘤肺转移率,其作用程度与内皮抑素剂量呈现明显正相关,且血管内皮抑素中未见获得性耐药现象发生。结论血管内皮抑素可以通过下调肿瘤组织中的MVD,抑制肿瘤生长及减少转移的发生,不会诱发肿瘤对化疗药物的耐药现象发生,值得在抗癌治疗中使用。
Objective To observe the effects of endostatin on Lewis lung carcinoma growth, angiogenesis and metastasis in mice. Methods C57 mice bearing Lew is lung cancer were treated with different doses of endostatin and endostatin. The expression of MVD and the incidence of metastasis in the tumor were observed. SPSS19.0 for statistical analysis. Results Endostatin significantly inhibited the growth of Lewis lung carcinoma in mice and significantly decreased the density of microvessels in the xenograft and metastatic tumors and decreased the lung metastasis rate. The effect of endostatin was positively correlated with the dose of endostatin No evidence of acquired resistance occurs in endostatin. Conclusions Endostatin can inhibit MVD in tumor tissue, inhibit tumor growth and reduce the incidence of metastasis, and will not induce tumor resistance to chemotherapeutic drugs. It is worth to be used in anti-cancer therapy.
