目的探讨膀胱肿瘤组织表达缺氧诱导因子 (HIF 1a)与肿瘤组织中微血管计数(MVC)、肿瘤分级、复发的关系。方法用免疫组化染色显示膀胱肿瘤组织中HIF 1a表达 ,并用计数定量血管形成 ,回顾性分析HIF 1a表达和MVC、肿瘤分级、复发的关系。结果正常膀胱粘膜 1 9例 ,膀胱肿瘤Ⅰ级 2 1例、Ⅱ级 2 7例、Ⅲ级 2 2例。HIF 1a表达 1 9例正常膀胱粘膜均为阴性 ,在Ⅰ、Ⅱ、Ⅲ级膀胱肿瘤中HIF 1a阳性率分别为 2 9%、41 %和 64 % ,Ⅰ、Ⅲ级阳性率差别显著 (P <0 0 5)。 1 9例正常膀胱粘膜MVC为 0 ,在Ⅰ、Ⅱ、Ⅲ级膀胱肿瘤中MVC分别为 39 71± 1 1 62、44 70± 1 1 52和 52 36± 1 6 83。其中Ⅰ、Ⅲ级相差显著 (P <0 0 5)。根据肿瘤属于初发或复发分组 ,初发组 51例 ,复发组 1 9例 ,HIF 1a阳性率分别为 35 %和 74% ,相差显著 (P <0 0 5)。MVC初发组和复发组分别为 42 68± 1 3 73和 53 47± 1 2 50 ,相差显著 (P <0 0 5)。结论HIF 1a阳性率和膀胱肿瘤MVC、肿瘤分级以及肿瘤复发率呈正相关关系 ,提示HIF 1a表达与膀胱肿瘤的生物学行为有关 ,可作为膀胱肿瘤生物学行为的标志
Objective To investigate the relationship between expression of hypoxia inducible factor (HIF 1a) and microvessel count (MVC) in tumor tissues, tumor grade and recurrence in bladder tumor. Methods Immunohistochemical staining was used to detect the expression of HIF 1a in bladder tumor. Quantitative angiogenesis was quantified by count. The relationship between HIF 1a expression and MVC, tumor grade and recurrence was retrospectively analyzed. Results Normal bladder mucosa was found in 19 cases, grade Ⅰ in 21 cases of bladder tumor, grade Ⅱ 27 cases and grade Ⅲ 22 cases. The positive expression of HIF 1a in grade I, II and III bladder cancer was 29%, 41% and 64%, respectively, and the positive rates of grade I and III were significantly different between HIF 1a and normal bladder mucosa (P < 0 0 5). The MVC of normal bladder mucosa was 0 in 9 cases, and 39 71 ± 1 1 62, 44 70 ± 1 1 52 and 52 36 ± 1 6 83 in grade Ⅰ, Ⅱ and Ⅲ bladder tumors respectively. The difference between Ⅰ and Ⅲ was significant (P <0 05). According to the group of tumors, the positive rate of HIF 1a was 35% and 74%, respectively, in 51 cases of new onset group and of recurrence group, the difference was significant (P <0.05). There were significant differences (P <0 05) between the MVC and the recurrent group for 42 68 ± 1 3 73 and 53 47 ± 1 50 respectively. Conclusions The positive rate of HIF 1a is positively correlated with MVC, tumor grade and recurrence rate of bladder tumor, suggesting that the expression of HIF 1a is related to the biological behavior of bladder tumor and may be used as a marker of biological behavior of bladder tumor
