目的 研究VP—16诱导雄激素非依赖型前列腺癌细胞PC—3凋亡和癌基因bcl—2及抑癌基因P53表达的关系。方法 以末端标记法(TUNEL)和流式细胞术(FCM)研究PC—3细胞凋亡及细胞周期分布,以免疫组化方法研究P53及bcl—2蛋白的表达。结果 末端标记法显示凋亡的PC—3细胞呈明显的形态学改变,流式细胞术和免疫组化检测结果表明,随VP—16作用浓度的增加和作用时间的延长,PC—3细胞的凋亡率和其P53基因表达阳性的细胞百分率逐渐增高,而bcl—2基因表达阳性的细胞百分率逐渐降低。结论 VP—16能够诱导PC一3细胞凋亡和增加P53基因表达及抑制bcl—2基因,且具有作用浓度和作用时间依赖性。
Objective To investigate the relationship between the apoptosis of PC-3 and the expression of bcl-2 and P53 in androgen-independent prostate cancer cells induced by VP-16. Methods Apoptosis and cell cycle distribution of PC-3 cells were studied by terminal labeling (TUNEL) and flow cytometry (FCM). The expression of P53 and bcl-2 protein was studied by immunohistochemistry. Results The end-labeling showed that the apoptotic PC-3 cells showed obvious morphological changes. The results of flow cytometry and immunohistochemistry showed that with the increase of the concentration of VP-16 and the prolongation of the action time of PC-3 cells. The percentage of cells with apoptotic rate and positive expression of P53 gene was gradually increased, while the percentage of cells with positive expression of bcl-2 gene was gradually decreased. Conclusion VP-16 can induce apoptosis of PC-3 cells and increase the expression of P53 gene and inhibit the expression of bcl-2 gene, and it has the effect concentration and action time dependence.
