目的探究紫杉醇对卵巢癌治疗效果以及对相关基因FOXM1表达影响。方法通过荧光定量PCR,Western blot方法从RNA以及蛋白质水平分别对卵巢癌患者卵巢标本(卵巢癌组)及因其他原因而行卵巢切除的卵巢标本(对照组)中FOXM1基因的表达进行测定,同时运用酶联免疫吸附法(ELISA)检测二者血清中FOXM1蛋白含量及卵巢癌患者服用紫杉醇后不同时间段(1、2、3、4、5、6月)血清FOXM1含量变化。结果卵巢癌患者卵巢中FOXM1基因在mRNA以及蛋白水平的表达量、血清中FOXM1含量均显著高于对照组(P<0.05);与治疗前相比,卵巢癌患者服用紫杉醇后血清中FOXM1含量随治疗时间的延长逐步降低(P<0.05),但治疗6个月后,仍显著高于对照组(P<0.05)。结论 FOXM1基因与卵巢癌发病之间存在密切关系,服用紫杉醇可以降低患者体内FOXM1基因表达量。
Objective To investigate the therapeutic effect of paclitaxel on ovarian cancer and its effect on the expression of FOXM1. Methods The expression of FOXM1 gene in ovarian cancer patients (ovarian cancer group) and ovariectomized ovarian specimens (control group) were detected by real-time quantitative PCR and Western blot respectively from RNA and protein levels. Meanwhile, Serum levels of FOXM1 protein in serum and levels of FOXM1 in ovarian cancer patients at different time points (1,2,3,4,5 and 6 months) after taking paclitaxel were detected by enzyme-linked immunosorbent assay (ELISA). Results Ovarian cancer FOXM1 gene mRNA and protein levels in ovarian expression, serum FOXM1 levels were significantly higher than the control group (P <0.05); compared with before treatment, ovarian cancer patients taking paclitaxel serum FOXM1 content The prolongation of treatment time gradually decreased (P <0.05), but after 6 months of treatment, it was still significantly higher than that of the control group (P <0.05). Conclusion There is a close relationship between FOXM1 gene and ovarian cancer. Taking paclitaxel can reduce FOXM1 gene expression in patients.
