以睾酮为模型药物,研究介质对药物经皮渗透的影响,测定了睾酮在不同介质中的溶解度及睾酮在这些介质中通过大鼠皮肤的渗透速率。经Hildebrand法计算得到药物和介质的溶解度参数。结果显示,睾酮的经皮渗透系数的对数(logp)与介质的溶解度参数[δv(n)]之间呈倒置抛物线相关,当介质的溶解度参数接近药物溶解度参数时药物的渗透系数达最小值。
Testosterone was used as a model drug to study the effect of media on transdermal drug penetration and to determine the solubility of testosterone in different media and the rate of testosterone penetration through rat skin in these media. The solubility parameters of drugs and media were calculated by Hildebrand method. The results showed that the logarithm of the transdermal permeability coefficient of testosterone was inversely parabolic correlated with the solubility parameter [δv (n)] of the medium. When the solubility parameter of the medium was close to the drug solubility parameter, the permeability coefficient of the drug reached the minimum value .
