目的:试从干预高血压病血管壁炎症细胞浸润角度,进一步完善复方钩藤降压片降压及防治靶器官损害的机制。方法:21只7周龄雄性自发性高血压大鼠(SHR)随机分为3组:模型组(SHR-S组)、复方钩藤降压片组(SHR-G组)、缬沙坦组(SHR-D组),另7只WKY大鼠为空白对照组(WKY组)。连续干预6周,HE染色观察腹主动脉白细胞黏附跨壁浸润情况、免疫组化法测定管壁E-选择素(Es)的表达水平。结果:(1)对腹主动脉壁白细胞黏附浸润的影响:SHR-S组管壁皆有不同程度白细胞黏附浸润,以黏附为主,内皮连续性较差,弹性纤维排列紊乱,平滑肌细胞显影模糊,部分大鼠平滑肌细胞有空泡样变性,SHR-D组管壁均有不同程度的白细胞黏附浸润,以中膜浸润为主,未见其他明显异常改变,WKY组及SHR-G组动脉管壁无上述异常病变;(2)对腹主动脉Es表达的影响:Es的表达水平SHR-G组低于SHR-S组(P<0.05),与WKY组与SHR-D组无显著差异(P>0.05),SHR-D组表达水平亦低于SHR-S组(P<0.05)。结论:SHR腹主动脉壁存在白细胞黏附浸润,且E-选择素表达水平升高,复方钩藤降压片能有效减少管壁白细胞黏附浸润及降低E-选择素的表达水平。
OBJECTIVE: To further improve the antihypertensive effect of compound Uncaria decoction and the mechanism of prevention and treatment of target organ damage from the perspective of intervention of inflammatory cell infiltration in the vascular wall of hypertension. Methods: Twenty-one seven-week-old male spontaneous hypertensive rats (SHR) were randomly divided into three groups: model group (SHR-S group), compound Uncaria decoction group (SHR- (SHR-D group) and the other 7 WKY rats as blank control group (WKY group). Continuous intervention for 6 weeks, HE staining of the abdominal aorta leukocyte adhesion transmembrane infiltration, immunohistochemical determination of E-selectin (E) expression. Results: (1) Effect on the adhesion and infiltration of leukocytes in the abdominal aorta wall: There were different degrees of leukocyte adhesion and invasion on the wall of SHR-S group, which were mainly adhered with poor continuity of the endothelium, disordered elastic fibers and fuzzy development of smooth muscle cells , Part of rat smooth muscle cells have vacuolar degeneration, SHR-D group had varying degrees of leukocyte adhesion infiltration, mainly in the infiltration of the media, no other significant abnormal changes in WKY group and SHR-G group arterial (2) The expression of Es on the abdominal aorta: The expression level of Es in SHR-G group was lower than that in SHR-S group (P <0.05), but not significantly different from WKY group and SHR-D group P> 0.05). The expression level of SHR-D group was also lower than that of SHR-S group (P <0.05). CONCLUSION: There is leukocyte adhesion infiltration in the abdominal aorta wall of SHR and the expression of E-selectin is increased. Compound Uncaria decoction can effectively reduce the adhesion and infiltration of tubular leukocytes and decrease the expression of E-selectin.
