目的建立高效毛细管电泳法(HPCE)同时测定甘草饮片中甘草次酸、异甘草苷、甘草苷、甘草素、甘草酸和异甘草素含量的方法。方法 40 mmol.L-1硼砂-10 mmol.L-1磷酸二氢钠-10%甲醇(pH 8.6)为电泳介质,未涂渍标准熔融石英毛细管(75μm×64.5 cm,有效长度56 cm)为分离通道,分离电压为20 kV,检测波长为254 nm,毛细管温度为20℃,压力进样为5 kPa×6 s。结果 6种指标成分的浓度与峰面积的线性关系良好(r>0.995 0);加样回收率为95.00%~100.90%;用此法测定了甘草饮片中6种指标成分的含量,结果满意。结论该方法简单、准确,重复性较好,可用于甘草饮片内在质量的评价和控制。
Objective To establish a method for simultaneous determination of glycyrrhetinic acid, isoliquiritin, glycyrrhizin, licorice, glycyrrhizinate and isoliquiritigenin in Glycyrrhiza glabra by high performance capillary electrophoresis (HPCE). Methods 40 mmol·L-1 borax-10 mmol.L-1 sodium dihydrogen phosphate-10% methanol (pH 8.6) was used as the electrophoresis medium without standard fused silica capillary (75 μm × 64.5 cm, effective length 56 cm) The channels were separated, the separation voltage was 20 kV, the detection wavelength was 254 nm, the capillary temperature was 20 ℃, and the pressure injection was 5 kPa × 6 s. Results The linear relationship between the concentration of 6 kinds of index components and the peak area was good (r> 0.995 0). The recoveries were 95.00% ~ 100.90%. The content of 6 kinds of index components in licorice root slices was determined by this method and the results were satisfactory. Conclusion The method is simple, accurate and reproducible. It can be used to evaluate and control the intrinsic quality of licorice root slices.
