目的探讨RASSF1A基因对人胃癌SGC7901细胞NF-κB活性的影响。方法电泳迁移实验分析RASSF1A基因转染前后SGC7901细胞NF-κB活性的变化。蛋白印迹和免疫细胞化学检测RASSF1A基因转染前后SGC7901细胞P65蛋白的表达。结果与未转染组和转染空白载体组比较,转染RASSF1A基因组SGC7901细胞的NF-κB活性明显降低,P65蛋白的表达明显减少。结论 RASSF1A基因可能通过抑制P65的表达、降低NF-κB活性抑制从而抑制胃癌SGC7901细胞生长。
Objective To investigate the effect of RASSF1A gene on the activity of NF-κB in human gastric cancer cell line SGC7901. Methods The electrophoretic mobility shift assay was used to analyze the changes of NF-κB activity in SGC7901 cells before and after RASSF1A gene transfection. Expression of P65 protein in SGC7901 cells before and after RASSF1A gene transfection by Western blotting and immunocytochemistry. Results Compared with the untransfected and transfected blank vector groups, the NF-κB activity of transfected SGC7901 cells was significantly decreased and the expression of P65 protein was significantly decreased. Conclusion RASSF1A gene may inhibit the growth of gastric cancer SGC7901 cells by inhibiting the expression of P65, inhibiting the inhibition of NF-κB activity.
