[目的]探讨鸟嘌呤酶(GUA)在肝细胞损伤过程中的动态改变及其临床价值。 [方法]以2-FAA喂饲SD大鼠,观察大鼠肝细胞从损伤到癌变过程中血清及肝组织中的GUA的活性变化。[结果]在鼠肝细胞发生变性至肝组织全数癌变时,各实验组血清及肝组织中GUA活性与正常对照组相比,GUA第二周开始升高,以后酶活性持续升高,但肝癌鼠GUA的动力学常数(Km值)与正常鼠的GUA相比,未见异常改变。[结论]胞浆酶GUA为肝细胞损害的特异标志酶,血清该酶活性的升高幅度反映了肝病的严重程度。
[Objective] To investigate the dynamic changes of guanine enzyme (GUA) in hepatocyte injury and its clinical value. [Methods] SD rats were fed with 2-FAA to observe the changes of the activity of GUA in the serum and liver tissues of rat hepatocytes from injury to carcinogenesis. [Results] When the rat hepatocytes were degenerated to complete hepatic carcinogenesis, the GUA activity in the serum and liver tissues of each experimental group was higher than that in the normal control group. The GUA began to increase in the second week, and the enzyme activity continued to increase, but the liver cancer increased. The kinetic constants (Km values) of murine GUA showed no abnormal changes compared with those of normal murine GUA. [Conclusion] The cytoplasmic enzyme GUA is a specific marker enzyme for hepatocyte damage, and the increase in serum enzyme activity reflects the severity of liver disease.
