目的 应用保尔佳 (Polyerga)体外净化自体骨髓移植治疗急性白血病及实体瘤 ,探索有效的体外净化新方法。方法 取正常骨髓加入不同浓度的保尔佳在 3 7℃条件下孵育后台盼蓝染色 ,计数活细胞数 ,细胞培养计数CFU -GM ,评价保尔佳对正常造血干细胞的影响。临床应用按 10 0~ 15 0ng/ml保尔佳体外骨髓净化处理 ,净化组预处理方案及回输自体骨髓细胞量与对照组相似。结果 不同浓度的保尔佳在体外对正常造血干细胞均无影响 ,净化组与对照组自体骨髓移植造血恢复时间相似 ,实验组与对照组间无差异 (P >0 0 5 )。随访报告提示 ,净化组复发率较对照组明显降低 (P <0 0 1)。结论 保尔佳体外净化自体骨髓对造血干细胞无损伤 ,可有效清除体内残留肿瘤细胞 ,延长无病生存期。
Objective To investigate the effective in vitro purification method of Polyerga in vitro autologous bone marrow transplantation for the treatment of acute leukemia and solid tumors. Methods Normal bone marrow was infiltrated with different concentrations of bovine trypan blue staining at 37 ℃, count the number of viable cells and count CFU-GM in cell culture to evaluate the effects of baojia on normal hematopoietic stem cells. Clinical application of 10 ~ 15 0ng / ml Paul in vitro bone marrow purification in vitro purification group pretreatment regimen and autologous bone marrow cells returned to the control group. Results Different concentrations of baojiajia had no effect on normal hematopoietic stem cells in vitro. The recovery time of hematopoietic cells in autologous bone marrow transplantation group was similar to that in control group. There was no significant difference between experimental group and control group (P> 0.05). Follow-up reports suggest that the recurrence rate of the purge group was significantly lower than that of the control group (P <0.01). Conclusion Purcell purification of autologous bone marrow in vitro does not damage hematopoietic stem cells, which can effectively remove residual tumor cells and prolong disease-free survival.
