目的:研究雌激素受体α(ER-α)基因PvuⅡ多态性与子宫内膜癌(EC)组织ER阳性表达、血清雌二醇(E2)水平3者间的相互关系。方法:选择60例EC患者为EC组和60例健康者为对照组,检测两组ER-α基因PvuⅡ多态性,检测EC组ER阳性表达及E2水平。结果:ER-α基因PvuⅡ多态性在EC组和对照组间差异有统计学意义(P<0.05),P等位基因患EC的风险是p等位基因的1.775倍(OR=1.775,95%CI:1.045~3.012);EC组PP基因型癌组织ER阳性表达明显少于Pp、pp基因型(P<0.05);在ER阳性表达的组织中,PP基因型ER阳性表达面积的百分比也明显0.05)。结论:ER-α基因PvuⅡ基因多态性与EC的发病有相关性,P等位基因可能是EC的遗传易感基因,PP基因型与癌组织ER阳性低表达具有关联性。
OBJECTIVE: To study the relationship between the Pvu Ⅱ polymorphism of estrogen receptor α (ER-α) gene and the ER expression and the level of serum estradiol (E2) in endometrial carcinoma (EC). Methods: 60 EC patients were selected as EC group and 60 healthy controls as control group. The Pvu Ⅱ polymorphism of ER-α gene was detected in both groups. ER expression and E2 level in EC group were detected. Results: The Pvu Ⅱ polymorphism of ER-α gene was significantly different between EC group and control group (P <0.05). The risk of EC in P allele was 1.775 times of that of the p allele (OR = 1.775, 95 % CI: 1.045-3.012). The positive expression of ER in PP genotypes in EC group was significantly less than that in Pp and pp genotypes (P <0.05). The percentage of ER positive expression areas in PP genotypes (P <0.05). There was no significant difference in serum E2 levels among the three genotypes of PP, Pp and Pp (P> 0.05). CONCLUSION: The polymorphism of PvuII gene of ER-α gene is correlated with the pathogenesis of EC. The P allele may be a genetic susceptibility gene of EC. The genotype of PP is associated with the low expression of ER in cancer tissue.
