单纯疱疹病毒1型(HSV-1)的gB是病毒囊膜上的几种糖蛋白之一。它决定着病毒的感染性,可能是宿主免疫应答的重要靶抗原之一。 实验分别用HSV-1KOS株和F株感染HEP-2细胞,提取mRNA,在体外网织红细胞溶解系统中翻译,并加入外源性膜(狗胰脏微粒体膜,且用~(35)S-蛋氨酸标记感染细胞和微粒体),用免疫沉淀实验测产物分子量。用抗gB-1单克隆抗体系和层析纯化得到gB—1。
The gB of herpes simplex virus type 1 (HSV-1) is one of several glycoproteins on the viral envelope. It determines the infectivity of the virus and may be one of the important target antigens of the host immune response. HEP-2 cells were infected with HSV-1 KOS strain and F strain respectively. MRNA was extracted and translated in vitro in reticulocyte lysate system. The exogenous membrane (membrane of canine pancreas microsome, - methionine-labeled cells and microsomes), and the molecular weight of the product was measured by immunoprecipitation. GB-1 was purified by chromatography with anti-gB-1 monoclonal antibody.
