通过对胡桃楸成花相关LFY基因进行研究,对缩短胡桃楸童期,促进其提早开花结实有着极为重要的意义。采取PCR扩增技术,在NCBI数据库中进行搜索,查找到与目的基因有关联的同源序列;并根据这些有关联的序列来进行引物设计,扩增目的基因全长。克隆LFY基因cDNA序列全长为1 297 bp,其中CDS序列长1 158 bp,编码385个氨基酸。NCBI数据库Blast比对表明胡桃楸LFY同源基因与其他物种的LFY同源基因同源性均在80%以上。将胡桃楸LFY同源基因命名为JmLFY(GenBank登录号:KX364241)。通过BioEdit软件和ProtParam ExPASy在线软件分析胡桃楸JmLFY蛋白质分子式为C_(1878)H_(2962)N_(566)O_(578)S_(13),分子量为43 134.4 Da;理论等电点pI值为6.78;不稳定指数为39.92,属于不稳定蛋白质。该研究为进一步探索LFY基因对胡桃楸成花的影响提供依据。
Studying the flower-related LFY gene of Juglans mandshurica has the extremely important significance to shorten the period of the Juglans mandshurica and promote its early flowering and fruiting. PCR amplification was used to search the NCBI database to find homologous sequences related to the target gene. Based on these related sequences, primers were designed to amplify the full length of the target gene. The full-length cDNA of the cloned LFY gene was 1 297 bp in length, of which the CDS sequence was 1 158 bp in length and encoded a protein of 385 amino acids. The Blast alignment of the NCBI database showed that the LFY homologous genes of Juglans mandshurica were more than 80% homologous to other LFY homologous genes. The Juglans mandshurica LFY homologous gene was named JmLFY (GenBank accession number: KX364241). The molecular structure of JmLFY was identified as C_ (1878) H_ (2962) N_ (566) O_ (578) S_ (13) by BioEdit software and ProtParam ExPASy online software. The molecular weight of JmLFY was 43 134.4 Da. The theoretical isoelectric point ; The instability index was 39.92, belonging to the unstable protein. The research provided the basis for further exploring the effect of LFY gene on flower formation of Juglans mandshurica.
