目的研究NO合剂(硝普钠、夹竹桃麻素、精氨酸)对糖尿病小鼠皮肤创面愈合的影响。方法50只雄性昆明小鼠腹腔注射链脲佐菌素以建立糖尿病小鼠模型,建模成功后按完全随机分组的方法分成对照组、L-精氨酸(L-Arg)治疗组、夹竹桃麻素(APO)治疗组、硝普钠(SNP)治疗组及NO合剂治疗组,每组10只小鼠。在小鼠背部建立创面,分别以相应药物创面下注射。创面动态摄像并于相应时间段取标本,观察创面组织愈合情况,计算创面愈合率;免疫组化染色检测创面成纤维细胞密度、胶原纤维面密度和血管密度。结果创面形成后第3天起,各药物治疗组小鼠创面愈合率较对照组明显上升(P<0.05,P<0.01),以创面形成后7 d内变化最为明显。与对照组相比,各药物治疗组小鼠的创面成纤维细胞密度、胶原面密度、血管密度均明显升高(P<0.05,P<0.01),其中NO合剂治疗组与对照组比较差异更显著,L-Arg、APO、SNP治疗组之间比较差异无统计学意义(P>0.05)。结论NO合剂可有效促进糖尿病小鼠创面愈合,较L-Arg、APO、SNP单独使用效果更为显著。
Objective To study the effects of NO (sodium nitroprusside, apocynin, arginine) on wound healing in diabetic mice. Methods Fifty male Kunming mice were injected intraperitoneally with streptozotocin to establish a diabetic mouse model. After successful modeling, the mice were randomly divided into control group, L-arginine group (L-Arg) (APO), sodium nitroprusside (SNP) and NO mixture treatment groups, 10 mice in each group. In the back of the mice to establish wounds, respectively, the corresponding drug wound injection. Wound dynamic imaging and samples taken at the appropriate time to observe wound tissue healing, wound healing rate was calculated; immunohistochemical staining wound fibroblast density, collagen fiber density and vascular density. Results From the third day after the wound was formed, the healing rate of the wound surface in each drug treatment group was significantly higher than that in the control group (P <0.05, P <0.01). The most obvious change occurred within 7 days after wound formation. Compared with the control group, the wound fibroblast density, collagen surface density and vascular density in each drug treatment group were significantly increased (P <0.05, P <0.01), and the difference between the NO mixture treatment group and the control group was more Significantly, there was no significant difference between L-Arg, APO and SNP treatment groups (P> 0.05). Conclusion NO mixture can effectively promote wound healing in diabetic mice, which is more effective than L-Arg, APO and SNP alone.
