目的探讨μ阿片受体基因(OPRM1)6个位点rs6912029、rs510769、rs3798676、rs7748401、rs589046和rs2075572单核苷酸多态性(Single nucleotide polymorphism,SNP)与海洛因成瘾者美沙酮维持治疗剂量的相关性。方法问卷调查收集696例美沙酮维持治疗门诊患者既往吸毒信息和目前治疗信息,结合多重PCR技术、Mass ARRAY i PLEX单碱基延伸技术和基质辅助激光解吸附电离飞行时间质谱分析技术对研究对象OPRM1基因6个位点进行SNP分型,探讨上述位点多态性与美沙酮维持治疗剂量间的关联。结果 OPRM1基因rs6912029、rs510769、rs3798676、rs7748401、rs589046和rs2075572共6个位点的等位基因、基因型和单体型频率在美沙酮治疗高剂量和低剂量两组间分布差异均无统计学意义(P>0.05)。结论尚未发现OPRM1基因6个SNP位点(rs6912029、rs510769、rs3798676、rs7748401、rs589046和rs2075572)与海洛因成瘾者美沙酮维持治疗剂量之间存在相关性。
Objective To investigate the relationship between the single nucleotide polymorphisms (SNPs) of rs6912029, rs510769, rs3798676, rs7748401, rs589046 and rs2075572 at the six positions of μ opioid receptor gene (OPRM1) and the doses of methadone maintenance therapy in heroin addicts Sex. Methods Questionnaires The past drug information and current treatment information of 696 methadone maintenance treatment outpatients were collected. Combined with multiplex PCR, Mass ARRAY i PLEX single base extension and matrix-assisted laser desorption / ionization time of flight mass spectrometry, the OPRM1 gene Six SNPs were genotyped to explore the relationship between polymorphism of these loci and dose of methadone maintenance therapy. Results The allele, genotype and haplotype frequency of OPRM1 gene rs6912029, rs510769, rs3798676, rs7748401, rs589046 and rs2075572 were not significantly different between the high and low dose groups of methadone treatment P> 0.05). Conclusion No association was found between the 6 SNP loci of OPRM1 gene (rs6912029, rs510769, rs3798676, rs7748401, rs589046 and rs2075572) and methadone maintenance dose for heroin addicts.
