目的 了解第二信使神经酰胺信号转导对鼻咽癌细胞生长的影响。方法 采用MTT法检测神经酰胺对鼻咽癌细胞株 (CNE 2 )增殖的抑制作用。光镜观察、荧光染色、流式细胞术检测细胞凋亡改变。Westernblot法检测p2 1WAF1的表达。结果 神经酰胺在 6 2 5、12 5、2 5、5 0 μm浓度下 ,对CNE 2细胞生长有明显的抑制作用 ,流式细胞仪检测发现亚G1峰出现 ,荧光染色可见核荧光强度增加、核片段化和凋亡小体。p2 1WAF1蛋白表达明显上调。结论 神经酰胺能诱导鼻咽癌细胞株CNE 2凋亡 ,且上调 p2 1WAF1蛋白表达。p2 1WAF1可能是神经酰胺诱导鼻咽癌细胞凋亡中起作用的因素之一。
Objective To understand the effect of second messenger ceramide signal transduction on the growth of nasopharyngeal carcinoma cells. Methods The inhibitory effect of ceramide on proliferation of nasopharyngeal carcinoma cell line (CNE 2 ) was detected by MTT assay. Light microscopy, fluorescence staining, and flow cytometry were used to detect changes in apoptosis. The expression of p2 1WAF1 was detected by Western blot. RESULTS: The ceramide significantly inhibited the growth of CNE 2 cells at the concentrations of 625, 125, 25, and 50 μm. Flow cytometry revealed that the sub-G1 peak appeared, and the fluorescence intensity showed an increase in the nuclear fluorescence intensity. Nuclear fragmentation and apoptotic bodies. P2 1WAF1 protein expression was significantly upregulated. Conclusion Ceramide can induce the apoptosis of nasopharyngeal carcinoma cell line CNE 2, and up-regulate the expression of p2 1WAF1 protein. P2 1WAF1 may be one of the factors that play a role in ceramide-induced apoptosis of nasopharyngeal carcinoma cells.
