DNA分子操作技术的发展,使生物学家能够在DNA水平上进行基因组差异的分析.RFLP是最近十多年发展起来的用于基因组研究的方法.它已被广泛应用于生物进化、起源、群体遗传等领域.但是对于那些高度自交,遗传背景狭窄的物种测难以有效地利用RFLP进行基因组分析.1990年Williams和Welsh同时开创了利用人工合成的短核苷酸引物经PCR反应进行基因组DNA多态性分析的新方法,即 RAPD技术(Random amplifiedPolymorphic DNAs).由于这一技术能快速、有效地进行基因组DNA多态性检测.因此虽然其诞生时间很短,但已广泛应用于基因组研究的各个方面.
