目的 检测胶质瘤中神经细胞粘附分子 (NCAM)和增殖细胞核抗原 (PCNA)的表达状况 ,以探讨不同病理类型胶质瘤中二者的表达及其相关性。方法 用抗NCAM及抗PCNA单抗对 8例正常脑组织和 5 1例胶质瘤进行免疫组化检测。结果 NCAM在正常脑组织中全部阳性表达 ,Ⅰ~Ⅱ级胶质瘤、Ⅲ级胶质瘤、Ⅳ级胶质瘤中NCAM阳性表达分别为 95 2 % (2 0 /2 1)、5 2 6 % (10 /19)、18 2 % (2 /11) ,阳性细胞含量随肿瘤恶性程度升高反而减少 ;NCAM阳性表达量与PCNA指数呈负相关 (r=- 0 6 7,P<0 0 1)。结论 脑胶质瘤中NCAM表达程度与胶质瘤的临床病理分级和细胞增殖活性呈负相关 ,它的低表达是高度恶性胶质瘤的标志之一
Objective To detect the expression of neural cell adhesion molecule (NCAM) and proliferating cell nuclear antigen (PCNA) in gliomas to investigate the expression and correlation of NCAM and PCNA in different pathological types of gliomas. Methods Eight cases of normal brain tissue and 51 cases of glioma were detected by immunohistochemistry using anti-NCAM and anti-PCNA monoclonal antibodies. Results NCAM was positively expressed in all normal brain tissues. The positive expression of NCAM in grade I-II glioma, grade III glioma, and grade IV glioma was 95 2 % (2 0 /21) and 5 2 6 respectively. % (10 /19), 18 2% (2 /11), the positive cell content decreased with the increase of tumor malignancy; NCAM positive expression was negatively correlated with PCNA index (r=-607, P<0 0 1). Conclusion The expression of NCAM in gliomas is negatively correlated with the clinical pathological grade and cell proliferation activity of gliomas. Its low expression is one of the hallmarks of highly malignant gliomas.
