目的:研究同种异体自然杀伤(natural killer,NK)细胞对人急性髓系白血病细胞株KG1A的杀伤率,探讨供者NK细胞表面杀伤细胞免疫球蛋白样受体2DL1(killer immunoglobulin-like receptor 2DL1,KIR2DL1)表达差异对NK细胞杀伤KG1A细胞活性的影响。方法:自9名健康志愿供者分离外周血NK细胞,流式细胞术检测NK细胞表面KIR2DL1的表达率,LDH释放法测定NK细胞在效靶比为20∶1时对KG1A细胞的杀伤活性。结果:NK细胞表面KIR2DL1的表达率存在个体差异,9名供者的表达范围为(33.20±1.95)%~(92.69±1.47)%;9名健康供者NK细胞对KG1A细胞的杀伤活性不同,杀伤率范围为(44.40±1.97)%~(93.70±1.12)%。不同个体NK细胞KIR2DL1表达率与其对KG1A细胞的杀伤率呈负相关(r=-1.00,P=0.00)。结论:不同个体NK细胞其表面KIR2DL1表达率与NK细胞对KG1A细胞的杀伤率呈负相关。
OBJECTIVE: To study the killing rate of allogeneic natural killer (NK) cells on human acute myeloid leukemia cell line KG1A and to investigate the effect of killer immunoglobulin-like receptor 2DL1 , KIR2DL1) expression on NK cell killing KG1A cell activity. Methods: Peripheral blood NK cells were isolated from 9 healthy volunteers. The expression of KIR2DL1 on NK cells was detected by flow cytometry. The cytotoxic activity of NK cells against KG1A cells was determined by LDH release assay. Results: The expression of KIR2DL1 on NK cells was different from that on NK cells. The expression range of KIR2DL1 from nine donors ranged from (33.20 ± 1.95)% to (92.69 ± 1.47)%. The cytotoxic activity of NK cells from nine healthy donors to KG1A cells was different, The killing rate ranged from (44.40 ± 1.97)% to (93.70 ± 1.12)%. The expression of KIR2DL1 in different NK cells was negatively correlated with the killing rate of KG1A cells (r = -1.00, P = 0.00). Conclusion: The expression rate of KIR2DL1 on NK cells of different individuals is negatively correlated with the killing rate of NK cells to KG1A cells.
