目的观察层粘连蛋白(LN)和Ⅳ型胶原(CoⅣ)在小鼠肾泌尿小管发育中的表达规律,探讨两者与肾泌尿小管发育的关系。方法选取胚龄(E)12、14、16、18d胎鼠和生后(N)3、7、14和40d仔鼠肾脏,应用免疫组织化学技术对小鼠肾泌尿小管LN和CoⅣ的表达进行定位观察;应用蛋白印迹技术对小鼠肾泌尿小管LN和CoⅣ的表达进行定量检测。结果 E12d,LN在输尿管芽上皮细胞基膜处开始微弱表达,而CoⅣ在输尿管芽无阳性表达;E14d,LN和CoⅣ在各期肾小管上皮细胞基膜处表达;E16d,LN和CoⅣ在集合管上皮细胞基膜处均有表达。随着肾脏的发育,LN和CoⅣ在肾泌尿小管的表达量逐渐增加。结论 LN和CoⅣ可能对小鼠肾泌尿小管发育和成熟起重要作用。
Objective To observe the expression of laminin (LN) and collagen Ⅳ (Co Ⅳ) in the development of renal tubules of mice and the relationship between them and the development of renal urinary tubules. Methods The kidneys of fetal rats and their postnatal (N) 3, 7, 14 and 40 d pups at embryonic day (E) 12, 14, 16 and 18 days were harvested. Immunohistochemistry was used to detect the expression of LN and Co Ⅳ Location observation; Western blotting was used to detect the expression of LN and CoⅣ in renal tubules of mice. Results E12d, LN began to weakly expressed in the basement membrane of the ureteric bud epithelial cells, whereas CoⅣ had no positive expression in the ureteric buds. At E14d, LN and CoⅣ were expressed in the basal lamina of renal tubular epithelial cells in each phase. At E16d, Epithelial basement membrane are expressed. With the development of the kidneys, the expression of LN and CoⅣ in the urinary tubules gradually increased. Conclusion LN and Co Ⅳ may play an important role in the development and maturation of mouse renal tubules.
