糖蛋白中痕量的完整寡糖链结构可用现代质谱方法测定。对寡糖的对氨基苯甲酸乙酯衍生物的液态二次离子质谱进行了研究,测定了基质效应,比较了正、负离子谱,使得麦芽七糖衍生物的最小检测量达到4pmol。应用氘标记类似物及高分辨质谱数据解释了IgM中N-连接的寡糖链分子结构。
Trace amounts of intact oligosaccharide chains in glycoproteins can be determined using modern mass spectrometry methods. Liquid secondary ion mass spectrometry of ethyl p-aminobenzoate oligosaccharides was studied, and the matrix effect was determined. The positive and negative ion spectra were compared, so that the minimum detectable amount of maltoheptaose derivatives reached 4 pmol. Deuterium-labeled analogues and high-resolution mass spectrometry data were used to explain the molecular structure of N-linked oligosaccharide chains in IgM.
