目的观察蜜橘皮中柠檬苦素总类似物对小白鼠胃排空率影响和对兔的离体肠管影响,为进一步研究提供药理学依据和临床用药指导。方法以吗丁啉组、蒸馏水组为对照组,柠檬苦素总类似物提取物分为高、中、低剂量组,观察4种溶液对正常白鼠胃排空功能的影响。以家兔肠管收缩强度为指标,观察高、中、低、3个剂量组(1g·mL-1,0.5g·mL-1,0.25g·mL-1)的柠檬苦素总类似物对兔离体肠管收缩的影响。结果高、低剂量(1g·mL-1,0.25g·mL-1)组均能促进小白鼠的胃排空率,低剂量组的排空率大于高剂量组和0.5mg·mL-1的吗丁啉组。高、中、低剂量组均能使兔离体肠管收缩的频率降低,低剂量组可以使兔离体肠管收缩的幅度增加,而高剂量组能使收缩幅度降低,并且随着浓度的增加,其振幅逐渐减小。结论不同浓度的柠檬苦素总类似物对小鼠的胃排空功能的形状呈现抛物线。即随着浓度的增加,排空率增大到一定程度后,又逐渐降低,对兔的离体肠管起双向调节作用。
Objective To observe the effect of limonin analogues on the gastric emptying rate in mice and the effect on the intestinal tract of rabbits in vitro, and to provide pharmacological evidence and clinical guidance for further research. Methods The domperidone group and distilled water group were used as the control group. The limonin total analogue extract was divided into high, medium and low dose groups. The effects of four solutions on gastric emptying function were observed. Intestine contractile strength of rabbits was used as an index to observe the effects of limonin total analogues in high, medium and low dose groups (1g · mL-1, 0.5g · mL-1, 0.25g · mL-1) Effects of isolated intestinal contractions. Results The gastric emptying rate of mice was promoted by high and low dose (1g · mL-1, 0.25g · mL-1) groups, the emptying rate of low dose group was higher than that of high dose group and 0.5mg · mL-1 Domperidone group. In the high, medium and low dose groups, the frequency of contractile contraction of the intestine in rabbits decreased. The contractile amplitude of isolated rabbit intestine increased in the low dose group, but decreased in the high dose group. With the increase of the concentration, Its amplitude gradually decreases. Conclusion Different concentrations of total limonoids analogues have a parabola shape on gastric emptying function in mice. That is, as the concentration increases, the evacuation rate increases to a certain extent, and then gradually decreases, the two-way regulation of the rabbit intestine from the role.
