目的探讨百事乐胶囊对慢性应激抑郁模型(CUMS)大鼠行为活动、学习记忆及海马显微结构和神经元凋亡的影响。方法将SD大鼠随机分为空白对照组、抑郁模型组、氟西汀组、百事乐胶囊高、中、低(2.88g/kg、1.44g/kg、0.72g/kg)剂量组,空白对照组外其它5组建立抑郁模型,造模同时ig给药,观察对大鼠Open-field、1%蔗糖偏食度、水迷宫的影响,HE染色观察模型大鼠海马结构的变化,免疫组化检测海马CA3区Bcl-2的表达;Western-blot检测海马Bcl-2的表达。结果百事乐高剂量组可提升模型大鼠水平及垂直活动次数(P﹤0.01),中剂量组可提升模型大鼠水平活动次数(P﹤0.05),高剂量组、中剂量组可提高模型大鼠的蔗糖偏食度(P﹤0.01或P﹤0.05),缩短模型大鼠的EL,并能明显升高模型大鼠穿越目标象限的次数;同时高剂量可缩短目标象限潜伏时间(P﹤0.05)。病理检测显示百事乐高、中剂量组可增加大鼠海马CA3区锥体细胞层厚度,促进海马神经元排列整齐、紧密;免疫组化结果显示百事乐高剂量组能升高模型大鼠海马CA3区BCl-2的表达(P﹤0.05)。Western-Blot显示百事乐高剂量能升高模型大鼠海马Bcl-2的蛋白表达(P﹤0.05)。结论百事乐胶囊能明显改善抑郁模型大鼠的抑郁行为及海马CA3区神经元结构的异常变化,并能减少海马神经元的凋亡。
Objective To investigate the effect of Bai Si Le capsule on behavioral behavior, learning and memory, hippocampal microscopic structure and neuronal apoptosis in rats with chronic stress depression (CUMS). Methods Sprague-Dawley rats were randomly divided into control group, depression model group, fluoxetine group, and Baishang capsule high, middle, and low (2.88g/kg, 1.44g/kg, 0.72g/kg) dose groups. Depression model was established in the other five groups except for ig administration. The effects of Open-field, 1% sucrose predilection, and water maze were observed. HE staining was used to observe the changes of hippocampal formation in model rats. Immunohistochemical detection was performed. The expression of Bcl-2 in hippocampal CA3 region and the expression of Bcl-2 in hippocampus by Western-blot. Results The Pepsi-high dose group could increase the number of horizontal and vertical activities of the model rats (P<0.01). The middle dose group could increase the number of horizontal activity of the model rats (P<0.05). The high-dose group and the middle-dose group could increase the model rats. The degree of sucrose predilection (P<0.01 or P<0.05) shortened the EL of model rats and significantly increased the number of times the model rats crossed the target quadrant; at the same time, the high dose reduced the target quadrant latency (P<0.05). Pathological examination showed that Pepsi-Gao, middle-dose group can increase the pyramidal cell layer thickness in hippocampal CA3 region of rats and promote hippocampal neurons to be arranged neatly and tightly; Immunohistochemistry results show that Pepsi-high dose group can increase BCl in hippocampal CA3 region of model rats. -2 expression (P < 0.05). Western-Blot showed that Pescoto high dose increased Bcl-2 protein expression in the hippocampus of model rats (P<0.05). Conclusion Baisle capsule can significantly improve the depression behavior and the abnormal changes of neuronal structure in hippocampal CA3 region in depressed rats, and it can also reduce the apoptosis of hippocampal neurons.
