目的探讨胚胎培养液在CO_2平衡及使用过程中pH的动态变化特性。方法应用血气分析仪检测人输卵管液(HTF)培养液在CO_2平衡过程中不同时间、CO_2浓度、液量、加油厚度以及失去CO_2支持后的二氧化碳分压(PCO_2)和pH的变化。结果胚胎培养液添加10%合成血清代用品(SSS)后,[HCO_3]降低2.19mmol/L,pH降低0.022。1.0ml培养液CO_2完全平衡时间为5h,加油后延长至15h,培养液经5.0%CO_2完全平衡的最终pH为7.270~7.280。培养液液面越高平衡效果越差,≥6.0ml培养液CO_2平衡15h后pH尚未稳定。CO_2浓度由5.0%提高到6.0%,培养液pH降低0.060。培养液加油越厚其平衡效果越差,矿物油预先经过CO_2平衡能明显提高平衡效果。失去CO_2支持的无油培养液在静止状况下pH稳定时间为23min,而加油培养液的pH稳定时间超过60min。结论胚胎培养液CO_2平衡宜采用适量多管和≥15h的过夜平衡方法;失去CO_2支持的无油平衡培养液的操作时间应尽量短,而加油培养液可稍延长;应尽量减少培养液的加油厚度,使用预先CO_2平衡的矿物油能明显提高平衡效果;应实测培养液pH来客观评价CO_2平衡效果或CO_2气体质量,选择适宜的CO_2气体浓度、平衡方式和平衡时间。
Objective To investigate the dynamic changes of pH in embryo culture medium during the balance of CO 2 and its use. Methods Blood gas analyzer was used to detect the changes of CO 2 concentration, volume of fluid, thickness of fueling and carbon dioxide partial pressure (PCO 2) and pH of human oviduct fluid (HTF) in different periods of CO 2 equilibrium. Results After adding 10% synthetic serum substitute (SSS) to embryo culture medium, [HCO 3] decreased 2.19 mmol / L, pH decreased 0.022.1.0 ml culture medium CO 2 complete equilibrium time was 5 h, extended to 15 h after refueling, The final equilibrium pH of% CO_2 was 7.270 ~ 7.280. The higher the liquid level of the culture fluid, the poorer the balance effect is, and the pH value of 6.0 ml of the culture medium is not stable after 15 hours of equilibrium. The CO 2 concentration increased from 5.0% to 6.0%, and the pH of the culture medium decreased 0.060. The thicker the refiner is, the poorer the balance effect is. The pre-balance of mineral oil can obviously improve the balance effect. The pH-stable time of the oil-free medium with CO 2 support at rest was 23 min, while the pH of the refueling medium was stable for more than 60 min. Conclusion The balance of CO_2 in embryo culture medium should be appropriate amount of multi-tube and overnight balance ≥15h; the operation time of oil-free equilibrium culture medium without CO_2 support should be as short as possible, while the refueling culture medium may be slightly prolonged; Thickness, the use of pre-CO 2 balanced mineral oil can significantly improve the balance effect; pH should be measured to evaluate the objective equilibrium effect of CO 2 or CO 2 gas quality, select the appropriate CO 2 gas concentration, equilibrium and balance time.
