目的:探讨microRNA-146a在口腔鳞状细胞癌(oral squamous cell caicinoma,OSCC)组织和细胞系中的表达并探讨其对OSCC细胞侵袭迁移能力的影响.方法:采用实时荧光定量PCR(Real-time PCR)检测60例OSCC组织和30例正常口腔黏膜组织中microRNA-146a的表达水平,并分析microRNA-146a的表达与OSCC临床病理学参数的关系.同时采用Real-time PCR检测microRNA-146a在人OSCC细胞系(SCC4、SCC15、Tca8113)和人正常口腔黏膜角化细胞系HOK中的表达水平,Transwell迁移和侵袭实验检测上调OSCC细胞microRNA-146a的表达后细胞侵袭和迁移能力的变化.结果:OSCC组织中microRNA-146a的表达水平明显低于正常口腔黏膜组织(P<0.05),并且microRNA-146a的表达与淋巴结转移和临床分期密切相关(P<0.05).OSCC细胞系中microRNA-146a的表达水平明显低于正常口腔黏膜角化细胞系(P<0.05).Transwell迁移和侵袭实验结果表明上调microRNA-146a的表达水平能够明显抑制Tca8113细胞的迁移和侵袭能力(P<0.05).结论:microRNA-146a在OSCC组织和细胞中表达减低,上调microRNA-146a的表达能够明显抑制癌细胞的迁移和侵袭能力,可能成为OSCC基因治疗的潜在靶点.“,”Objective:To evaluate the expression of microRNA-146a in oral squamous cell carcinoma(OSCC)tis-sues and cell lines,and to assess the effect of microRNA-146a on migration and invasion in OSCC cells. Methods:Quantita-tive real-time PCR(qRT-PCR)was used to assess the expression levels of microRNA-146a in 60 cases of OSCC tissues as well as 30 cases of normal oral mucosa tissues,followed by analyses on the correlation between microRNA-146a expression and the clinicopathologic parameters of OSCCs. Meanwhile,the expression levels of microRNA-146a in human OSCC cell lines(SCC4,SCC15,Tca8113)andnormalhuman oral keratinocyte line(HOK)were detected by qRT-PCR. In addition,the migration and invasion abilities of OSCC cells after up-regulating microRNA-146a were assessed by Transwell migration and invasion assays. Results:The microRNA-146a expression was significantly lower in OSCC tissues compared to that in normal oral mucosa tissues(P<0. 05),and the microRNA-146a expression was significantly associated with lymph node metastasis and clinical stage(P<0. 05). Moreover,the microRNA-146a expression was significantly lower in OSCC cell lines than that in normal oral keratinocyte line(P<0. 05). Transwell migration and invasion assays demonstrated that migration and invasion abilities of Tca8113 cell was significantly suppressed by up-regulating microRNA-146a expression(P<0. 05). Conclusion:The expression of microRNA-146a was decreased in OSCC tissues and cells. The migration and invasion abilities of OSCC cells were significantly inhibited by up-regulating microRNA-146a,which was a potential targetingene therapy of OSCC.
