Cell membrane calcium (Ca2 +) transport disorder and the incidence of hypertension are closely related. The authors recently established a method of calcium uptake of the red blood cell outside of the vesicle (IOV) and provided a powerful tool for studying the abnormal Ca ~ (2+) uptake mechanism in hypertensive patients. Erythrocytes were first ruptured with hypotonic buffer (10 mmol / L Tris-HCl, pH 7.6-7.8) and repeated three times until a hemoglobin-depleted red blood cell vesicle (RBC-Ghost, milky white) was obtained. The cells were then incubated in a hypotonic, alkaline environment (0.5 mmol / L Tris-Hcl + 50 μmol β-mercaptoethanol + 20 μmol / L Tris-EDTA, pH 8.5)
