目的用氯胺T法标记眼镜蛇神经毒素(Neurotoxin,NTX),观察其在大鼠体内的分布。方法实验组和对照组的大鼠分别单次静脉注射125I-NTX50μg.kg-1和相同剂量的Na125I与NTX的临时混合液,于给药后30min和2h分两批处死,取各脏器组织样本称重并计算每g组织的cpm。用组织与血液的脉冲比值作为放射性相对掺入量的标准,用实验组(e)和对照组(c)相对放射性掺入量的比值(e/c)作为判断125I-NTX在大鼠各脏器组织分布多少的依据。结果大鼠静脉给125I-NTX50μg.kg-1后30min,分布最多的是肾脏,每g组织放射性达44cpm,为对照组的11倍。膀胱及尿、肺、肠及内容物、肾上腺亦有较高分布。给药后2h,分布最多的仍是肾脏,但比0.5h时减少一半左右。结论NTX主要分布在肾脏,肾脏以外的实质性脏器分布均较少,脑中分布最低。
Objective To investigate the distribution of Neurotoxin (NTX) by chloramine T method in rats. Methods Rats in experimental group and control group were given a single intravenous injection of 125I-NTX 50μg.kg-1 and the same dose of Na125I and NTX mixed solution at the same time. After 30min and 2h, The sample is weighed and the cpm per g of tissue calculated. The pulse ratio of tissue to blood was taken as the standard of relative radioactivity incorporation, and the ratio of radioactive incorporation (e / c) of experimental group (e) and control group (c) Organizations based on how much tissue distribution. Results After intravenous injection of 125I-NTX at 50μg.kg-1 for 30min, the kidney was the most distributed in rats. The radioactivity was 44cpm per g tissue, which was 11 times of the control. Bladder and urine, lung, intestine and contents, adrenal also have a higher distribution. 2h after administration, the most distributed kidney is still, but it is reduced by about half compared with 0.5h. Conclusion NTX mainly distributes in the kidney, while the distribution of the other organs outside the kidney is less, and the distribution in the brain is the lowest.
