目的 对人源汉坦病毒疫苗候选株作分离鉴定及特性的研究。方法 用Vero细胞分离病毒 ,用免疫荧光法作病毒效价测定。用RT PCR及部分核苷酸序列分析等检定。结果 用Vero细胞从安徽省凤台县肾综合征出血热病人血清中分离一株汉坦病毒 (HV) ,其抗原特性及PCR分型结果证明为汉滩病毒 (Ⅰ型病毒 ) ;其免疫血清对来源于不同地区的HV均有中和活性。M片段部分序列分析结果表明 ,该毒株与 76 118等毒株核苷酸序列有较大差异 ,但氨基酸序列同源性甚高。病毒株对Vero细胞敏感 ,繁殖高峰一般在 9天以后 ,可多次收获病毒。结论 上述特性表明 ,分离病毒株适合用作研制灭活疫苗后备株。
Objective To isolate and identify human Hantavirus vaccine candidate strains and study their characteristics. Methods Vero cells were used to isolate the virus and immunofluorescence was used to determine the virus titer. RT PCR and partial nucleotide sequence analysis and other tests. Results Hantavirus (HV) was isolated from the serum of patients with HSV in Fengtai County of Anhui Province by Vero cells. The antigenicity and PCR typing results showed that it was Hantaan virus (type I virus) HV from different regions have neutralizing activity. Partial sequence analysis of M fragment showed that there was a great difference in nucleotide sequence between the strain and strain 76 118, but the amino acid sequence homology was very high. Virus strains sensitive to Vero cells, breeding peak is usually 9 days later, the virus can be harvested several times. Conclusion The above characteristics show that the isolated virus strains are suitable for the development of inactivated vaccine reserve.
